his is an interesting question. I suppose it depends in part on the point at which he loses.
I think if he somehow loses the Republican nomination, his response will be different than if he loses the general election. My guess (and all of this is just speculation) is that if he loses the nomination, after leading in the polls for a year and winning a lot of early primaries, he will be angry, will accuse the party of stealing the nomination from him, and do what he can to sink their chances of winning the general election. It is very hard to imagine him supporting the eventual Republican nominee if, for example, there is a brokered convention in which Trump has more delegates than anyone else (but not a majority) and still walks away without the nomination. This is the most likely scenario whereby he'd lose the nomination at this point, and he will almost certainly see this as a kind of cheating being used against him. He's got the support of about 1/3 of the Republican electorate, and he can use that to do some real damage if the nomination is taken from him, either by mounting a 3rd party challenge (which would be difficult, but not impossible) or otherwise sabotaging the Republican nominee, perhaps by encouraging his own supporters to stay home.
But, if he IS the Republican nominee, and loses the general election, it will be interesting to see how he reacts. There is a protocol to how losing candidates are supposed to behave. Once it is clear, on election night, that they have lost, they are expected to call the winner, congratulate them, and concede the race. This is not a requirement and has no impact on the outcome of the election, but it's the normal protocol. Then, they usually give a concession speech in front of their key supporters and campaign staff, and the expectation is that they call for unity now that the election is over. Finally, the losing candidate is expected to accept that they will now fade from public view, as attention shifts to the new president-elect.
If Trump loses, it will be interesting to see how he acts. Will he concede on election night? Will he give a concession speech, and if so, will it be conciliatory and humble, or will he rage and lash out angrily? And, perhaps most importantly, given his ability to command media attention, will he fade away in the days after the election?
These are all good questions. I wish I could tell you I have the answers, but Trump is wildly unpredictable. So at the very least, it would make for an interesting election night.

Whether Trump wins or loses, his supporters will not go away. I'm guessing there are about 15% of Americans who are crave authoritarian rule, which is why they are Trump supporters despite the insanity. The means don't matter to them--they are willing to be indecent, racist, xenophobic, violent, dividers. These supporters will not go away if Trump loses. They might submerge for a while, waiting for their next opportunity.

Perhaps it should be “when” he looses. I saw a statement by someone long involved in the Republican party suggesting that he is the Emperor who has no clothes. That seems apt from what I have observed of him. (I have tried commercial dispute cases for decades and I have negotiated commercial matters around the world, so that my impression of Mr. Trump are based upon that paradigm.)
He can present statements that are clearly on their face un-maintainable under cross examination, but which he seems to think are completely valid. Thus, if he were to loose, and he has not come to that resolution internally by the time it occurs, there are no limits as to what he might do.
As the 50 Republicans recently pointed out, his character make up does not allow for any fault on his part in what he undertakes and he will be like a little kid in his reaction. The problem will not be with him, but with a minority of his supporters, as they seek to undo the injustice that they think will have occurred. The Trump Card has no appreciation for the harm he could do to the entire Constitutional model, which has been exceptional for about 232 years.

I don’t think I’ve ever heard someone applying for a job say they’ll take a vacation if they don’t get the job. It’s practically like giving up in political jargon.
The correct answer should have been: “I’m looking towards the future, and thinking about what we can do to make America great and blah blah blah. I’m putting my faith in the American people.”
